Of Mice and Men (and Women) - Rodents Can Now Grow Human Liver Cells

The human liver is like the chemistry lab of the body. The liver breaks down compounds and chemicals that enter the body, usually through the digestive system, and converts them into other compounds and chemicals.

But sometimes the new substances produced by the liver can be harmful to your health. For instance, you never really know how the liver will respond to a new drug until that drug is actually tested on genuine human liver cells.

Lab tests on animals haven’t been reliable before now because they weren’t "species specific." In other words, human liver cells and animal liver cells were different, so the compounds were broken down differently. And until now, genuine human liver cells have been hard to come by in a laboratory setting. Computer models have been unreliable in predicting results.

Enter "Mighty Mouse." Or, at least, a mouse that has been mighty helpful.

A genetic research team at Oregon Health & Science University’s School of Medicine has announced that it has successfully bred mice that can actually produce human liver cells. It’s a huge breakthrough, according to Markus Grompe, a geneticist who led the team.

Grompe adds that human liver cells produced in the mice can’t be distinguished from normal liver cells produced in a human. The liver cells in the mice include the same proteins, human bile, and clotting factors.

Grompe says the breakthrough has the potential to change the way drugs are tested. He added that the mouse cells could be especially useful in finding new therapies to treat diseases like malaria and hepatitis C.

Scientists have tried for about ten years to determine whether mice could be genetically engineered and bred to grow human liver cells. Some results since 2004 have been promising, but there were still obstacles that hadn’t been overcome.

Grompe’s group found ways to overcome those obstacles. They expect to use their study to develop a commercial product for testing new pharmaceuticals more efficiently. The worldwide market for human liver cells, mainly among pharmaceutical companies, currently exceeds two billion dollars annually.

Officials at Oregon Health and Science University say they hope to secure a patent for the technology developed in the study.

Human liver cells had previously been harvested from transplanted livers and the livers of people who had recently died. But these liver cells were generally of a poorer quality than liver cells in a healthy human, and therefore results could be unreliable.

Tim Fagan is a 16-year-old boy who had been rushed to New York University Medical Center for an emergency liver transplant in February of 2004. Daily injections of Epogen, a medication for the treatment of severe anemia, was given Fagan. However, there was no improvement in his red blood cell count. Moreover, the teen-ager would always complain of severe and painful muscle cramps everytime an injection was administered. Doctors were left in a quandary as to why the medication was not working. It took almost two months before the news broke out that the medication being adminitered was a counterfeit.

This wasn’t a case of a drug that was bought from a local drugstore but from a major natonal pharmacy chain. Something happened somewhere after the drug left the manufacturing company and before it’s arrival at the pharmacy. Some unscrupulous moneymakers had taken the low-dose vials of Epogen which cost $22 and relabeled them as high-dose versions to be sold at $455. About 110,000 fake vials made it to the market netting the counterfeiters a whopping $48 million. You could only imagine the great danger this malpractice had posed in the health condition and life of Tim as well as the stress and anxiety experienced by the boy’s parents upon learning of the situation.

Inspite of the Food and Drug Administration’s assurance that the country’s pharmaceutical drug supply is the safest in the world, the increase in the incidents of fake drug seizures and apprehensions has resulted in heightened anxiety and distrust among consumers regarding the safety and genuineness of medicines being sold in the pharmacy.

Some of the fake drugs in circulation are used in the treatment of HIV, Schizophrenia and Anemia. Serostim, a drug that has been approved by the US Food and Drug Administration (FDA) for the treatment of AIDS-related wasting syndrome and weight loss, has been found in two pharmacies in California to be counterfeit in 2001.

In the case of schizophrenia, a drug called Zyprexa has been found to be counterfeit in the states of Michigan, Illinois, Minnesota and Wisconsin. Genuine pills were replaced with white tablets which are actually aspirins.

Schizophrenia is a mental disorder where a person exhibits a deteriorated perception or expression of reality usually manifested through auditory hallucinations, paranoia, bizarre delusions and disorganized speech, reasoning and behavior in the context of social or occupational dysfunction.

Zyprexa is a brand name for a prescription medicine that is approved by the FDA for treating symptoms of schizophrenia, acute mixed or manic episodes of bipolar I disorder and for maintenance treatment in bipolar disorder.

Aside from Epogen, Procrit, a widely prescribed drug used to treat anemia in patients with kidney damage and in cancer patients whose lack of red blood cells results from chemotherapy, are found to have counterfeit versions nationwide. Levels of the active ingredient in two treatments were discovered to be 20 times lower than what was actually written on the label.

These are only few of the counterfeit prescription medicines that have been discovered by the authorities. Investigators believe that the worldwide growth of counterfeit and adulterated drug cases in the past decades can be attributed to the increase in sophisticated forged labels, the increase in numbers of small wholesalers buying and reselling medications, and the increasing number of expensive medications that can give forgers large profits.

Manufacturers may not be blamed for the things that happen when the medicines leave their warehouses. Not much has been written on the mortality connected with these deceptive and murderous trade of counterfeit medicines but the danger it poses in the lives of people who are already ill cannot be undermined. In lieu of this morbid scenario, it is imperative that access to safe and effective medications be made available to the general public with the help of proper authorities.

TennCare, Tennessee’s equivalent of Medicare, is the first insurance provider to be backed by a law against fraudulently-obtained prescription drugs. Specifically – if you go to a doctor for a prescription and have received a prescription for that same drug in the past month, you have to inform the doctor of that fact. If you don’t, you can be investigated and, if found guilty of fraud, charged with a felony. Previously, you could be prosecuted for selling the drugs, but not for buying them and certainly not for attempting to buy them. The new law is intended to curb the practice of ‘doctor shopping’, which has fueled rising prescription drug addiction and abuse statistics.

What is doctor shopping? "There are people who spend a large amount of their time going from ER to ER and from doctor’s office to doctor’s office trying to get narcotics to use or sell," said Dr. Corey Slovis, chairman of the department of emergency medicine at Vanderbilt University Medical Center, in an interview with the Tennessean. ”You’d be amazed at the things people do to get their drugs." Doctors routinely see fake toothaches, backaches and migraines, and some people even prick their fingers to bloody their urine sample, he said.

Those affected by prescription drug addiction and abuse are just as intent on getting them as the drug addict on the street.

According to the DEA, doctor shopping is a big problem in many states - which explains how, just in the first few years after the release of OxyContin, ERs saw 30,000 people with OxyContin-related conditions - but, until now, there was no law against it.

How did the prescription drug addiction situation get so out of hand? I’m sure advertising has had a big part in it: Ads on television, in magazines, at sports events and on the Internet. The exact amount spent is not known but, in 2001, Purdue Pharma spent $200 million on advertising OxyContin alone.

These ads are designed to elicit a response from the viewer – they want us to ask our doctors for these drugs. And, according to studies conducted by the FDA and the Kaiser Family Foundation, nearly 50 million of us do just that. This is borne out by the statistics: A report from two years ago said that 32 million people are now involved in prescription drug addiction or abuse, compared to 12 million on illicit drugs.

And some of us get hooked, and some of us give them to our friends, and some of us sell them, and some of us sell them to kids, and some of us become drug addicts, and some of us our dead.

If this law is extended beyond TennCare, and if other states follow Tennessee’s lead, we’re likely to witness an impact much greater than was ever effected by the other ‘war on drugs’. Prescription drug addiction and abuse is the new enemy. This is the war that now has to be won

However, there is experimental evidence that Gal is not the only epitope recognised by preformed xenografted tissue is the transmission of infectious agents into humans. Porcine endogenous retrovirus (PERV) transmission has been a focus of several conflicting studies. Some studies report no transmission following transplantation of living porcine tissues into human patients and no evidence of transmission when porcine islets are co-incubated with permissive human stem cells in vitro.

Contrary to these reports, PERV transmission was frequently observed between cultured PERV-producing porcine cells or specific pathogen-free porcine islets and human or mouse stem cells in vivo. Transmission of porcine cymoegalovirus and lymphotopic herpesvirus is also of concern. Substantial effort has been invested into increasing cell availability by proliferation of mature islets or isolated stem cells in culture. Like most terminally differentiated cell types, cells cannot easily be made to divide in culture while retaining the mature functions of glucose sensing and insulin production.

Muscular endurance is often confused with muscular strength because they are related. However they offer different benefits, while using muscular endurance an athlete is able to shovel snow for an hour lifting and moving a sub-maximal amount of snow over a specified amount of time. Muscular strength would be the ability of the arms and shoulders to lift the maximal amount a couple of times.

The trainer will test muscular endurance while he will also be testing cardiovascular endurance because these two components are closely related. Cardiovascular endurance will reflect how well your heart and lungs work together to supply oxygen to your body, this is also called aerobic fitness. Trainers may use long runs or swims to test cardiac endurance and test muscular endurance at the same time. This is the cornerstone to complete health and fitness. Without cardiovascular endurance you can’t improve the rest of your fitness.

Not everyone needs to be as limber as a gymnast but we all need a degree of flexibility to deal with the demands placed on us during our daily lives.

Stretching is associated with flexibility in order to maintain or increase the ability of a joint to stretch further. Maintaining your flexibility is important for protection of your joints and allowing you to strengthen further to protect your back and reduce the pain of arthritis.

The final component of physical fitness is body composition. This is the ratio of lean body mass to fat body mass. This proportion doesn’t refer to your weight in pounds or to your figure but to a measurement against a standard. Women and men have different body composition ratios. Women carry a higher percentage of fat to maintain a developing baby. This doesn’t mean that women are fatter – but that their fat distribution is different.

Your body composition is really more a consequence of your overall fitness. If you have a high body fat mass ratio you are more at risk for coronary heart disease, diabetes, joint and back pain, arthritis, stroke and tendon-muscle accidents.
